Description

Half-Dye Mix reduces the costs involved in auto-sequencing, by reducing the amount of ABI™ BigDye Terminator needed in a reaction. Auto-sequencing reactions can leave up to 80% of dye terminators unused, which normally require removal prior to sequencing.

Half-Dye Mix works by providing optimal buffer conditions, which allow up to a 5-fold dilution of dye terminator without any loss of sequencing quality. Half-Dye Mix is suitable for sequencing of plasmid and PCR templates, and requires no optimization. For some templates it may be necessary to adjust the dilution factor.

Protocol for PCR Templates:

For PCR templates at a concentration of at least 20ng/μl, Dye-terminator can be diluted 5 fold with Half-Dye Mix.

In a normal 20μl reaction the dye-terminator accounts for 8μl.

The following mix is recommended: Big-DyeTM terminator ready reaction mix: 1.6μl
Half-Dye mix: 6.4μl
Total: 8.0μl

Add template and primers as per standard sequencing protocol and proceed as normal.

Protocol for Plasmid Templates:

For Plasmid templates at a concentration of at least 50ng/μl dye-terminator can be diluted 3 fold with Half-Dye Mix.

In a normal 20μl reaction the dye-terminator accounts for 8μl.

Using half-dye the following mix is recommended: Big-DyeTM terminator ready reaction mix: 2.6μl
Half-Dye mix: 5.4μl
Total: 8.0μl

Add template and primers as per standard sequencing protocol and proceed as normal.
